Abstract:A Gut-on-Chip is a microfluidic device containing intestinal cells that aims to better resemble the physiology of the intestine that a conventional cell culture on a wells plate can. A Gut-on-Chip design was already made for a poly(dimethylsiloxane) (PDMS) chip which contained eight culture chambers that are individually addressable by actuating Quake valves in flow channels before and after them. The aim of this research was to create a multiplexed Gut-on-Chip containing uniform cell layers in the individually addressable culture chambers of this chip. Coatings of collagen type I and poly(Llysine)poly(ethylene glycol) (PLL-g-PEG) were tested on cell adhesion. Furthermore two layers were made thinner to ensure proper valve closure. The control layer differs in thickness in a chip and between multiple chips and the chip did not have a lifespan longer than a few hours. The chip therefore still has problems when used for long-term experiments. An initial culture experiment of 18 hours was however performed on the multiplexed chip. Caco-2 cells were cultured in the culture chambers and only adhered in the desired places. The chambers were individually addressable and with some improvements, the chip has great potential to be used as a multiplexed Gut-on-Chip.
